ShrimProtect RxReady AHPND qPCR Kit has been validated by Balai Pengujian Ikan & Lingkungan (BPKIL) and has acquired certificates NIE KKP RI D 2212684 BBC to specifically detect pirA and pirB genes of AHPND/EMS-causing Vibrio parahaemolyticus (VPAHPND). The kit also contains Internal Control for shrimp.
Procedures
Store the reagents at -20oC upon delivery.
When ready to use, thaw completely and pipette 15 µL to each well of the PCR tube strip or well plate.
Mix 5 µL of the purified DNA and you are ready to run the PCR reaction.

Which organs or tissue that are recommended for use in AHPND PCR test?
Samples of gut-associated tissues and organs, such as hepatopancreas, stomach, the midgut and the hindgut are suitable for testing. In addition, faecal (non-lethal) samples may be collected from valuable broodstock.

What is the difference between pirA and pirB and the correlation with the occurrence of AHPND in shrimp?
The pirA protein play an auxiliary role in the pathogenesis of AHPND, while pirB alone could cause cellular damage, such as necrosis, apoptosis and other pathophysiological damage associated with AHPND.
How to interpret AHPND positive in the samples?
Samples are positive if there is amplification of pirA and/or pirB genes and it is positive as long as the Ct value is under 40.
